Dopamine-Synthesizing Neurons: An Overview of Their Development and Application for Cell Therapy

Cell-gene therapy is a dynamic constituent of novel medical biotechnology. Neurodegenerative disordersin which damage to or demise of specific brain cell types plays central role, are clear examples of diseasecandidate for cell replacement therapy. Dopaminergic (DAergic) neurons biosynthesize dopamine, a vitalneurotransmitter in the central nervous system. Due to the involveme...

Alpha-Synuclein Disrupted Dopamine Homeostasis Leads to Dopaminergic Neuron Degeneration in Caenorhabditis elegans

Disruption of dopamine homeostasis may lead to dopaminergic neuron degeneration, a proposed explanation for the specific vulnerability of dopaminergic neurons in Parkinson's disease. While expression of human alpha-synuclein in C. elegans results in dopaminergic neuron degeneration, the effects of alpha-synuclein on dopamine homeostasis and its contribution to dopaminergic neuron degeneration i...

Effects of Intranasally Administered Dnsp-11 on the Central Dopamine System of Normal and Parkinsonian Fischer 344 Rats

OF DISSERTATION EFFECTS OF INTRANASALLY ADMINISTERED DNSP-11 ON THE CENTRAL DOPAMINE SYSTEM OF NORMAL AND PARKINSONIAN FISCHER 344 RATS Due to the blood-brain barrier, delivery of many drugs to the brain has required intracranial surgery which is prone to complication. Here we show that Dopamine Neuron Stimulating Peptide 11 (DNSP-11), following non-invasive intranasal administration, protects ...

Long-term K+ channel-mediated dampening of dopamine neuron excitability by the antipsychotic drug haloperidol.

Antipsychotic drugs require days of treatment to begin to produce therapeutic effects. We report that in vivo treatment with the antipsychotic drug haloperidol acts with a delay of days to slow spontaneous repetitive firing by isolated midbrain dopamine neurons. The decreased excitability is caused by an increased number of functional A-type K+ channels without any change in gating properties. ...
